The Protein Source Station

The protein source station is an area in the software where you interact with your protein source. The NT8 will automatically recognize whether you attach a High Volume or Low Volume protein tray to the NT8 protein deck station. Here you define the initial protein source volume and the protein liquid class. These settings are needed for sitting drop, hanging drop, seeding, and serial dilution experiments.

Setting the Protein Volume

Prerequisites

 

To set the protein volume:

  1. If you are using a LV protein block, right-click the protein source image and select Set Protein Volume. If you are using a HV protein block, select Set High Volume Protein Tray. The Protein Volume Setup dialog appears:

  1. Leave the box next to Auto checked. If it is unchecked, the NT8 will not automatically use the reagent liquid class as defined in the Task Details dialog box.
  2. Fill in the volume available in each reservoir of a particular column. If you are using a high volume protein block, you can specify a volume of up to 13 μL.

Important: Please make sure that you set the volume with the actual volume available in each reservoir. You must have the same volume in each reservoir across a column. The column number should match the actual column number as the protein source that has been filled with protein.

  1. Click OK.

Aspirate/Dispense

  • Action: Provides two options: Aspirate and Dispense. Select Aspirate if you want the NT8 to aspirate protein from the protein source.
  • Column: Enables you to select the protein source column number. The NT8 will perform the action on the selected column. Please note that the protein column numbers will be disabled if you haven't set the protein volume (see Setting the Protein Volume).
  • Volume: Set the protein volume to be aspirated or dispensed by typing a new value into the corresponding field, or clicking the up and down-arrows to increase or decrease the volume.
  • Liquid Class: Enables you to select the protein liquid class.
  • Add: Adds the specified settings to the Task List and closes the Task Details dialog.
  • Add More: Adds the task to the task list and waits for you to add another aspirate or dispense command.
  • Close: Exits the Task Details dialog.

Note: The Aspirate/Dispense option is enabled only if you have already added the Pick Tip command to the Task List (see The Tip Caddy Stations).

Use Custom Aspirate

Displays the Aspirate Custom Height menu.

Important: This option is for advanced users or support only!

Set Protein Volume

See Setting the Protein Volume.
Set High Volume Protein Tray/Set Low Volume Protein trayEnables you to select the specific protein tray type to be used in your experiment. The NT8 supports the use of low volume or high volume protein trays.
