If automatic volume tracking isn’t necessary, users can opt to directly create and execute a sequence of cycles using MANUAL PROFILE. This can be used for slower concentrations that will run for a significant amount of time making volume tracking unnecessary, or when doing repetitive buffer exchange steps. It includes the same seven cycles available in a profile: Priming, Concentrating, Buffer Exchange, Air Recovery, Buffer Recovery, Tube Mixing, and Recirculation.

µPULSE allows you to create and run a sequence based on the number of pumps and time (in minutes) available in the MANUAL PROFILE menu. Follow these steps to set up a new profile:
µPULSE Home Screen
Tap the button.
Tap on the cycle card and select one of the cycles according to your preference.
Configure your cycle settings from the following options.
ON/OFF button to include or skip the step in the designed sequence.
Select between the number of pumps or the number of minutes for which you may want to perform that operation.
Selecting the ON/OFF Switch Button
Tap on the button to open the pressure settings and adjust the required pressures according to the sequence requirements.
Manual Profile Settings
Tap SAVE after adjusting the pressure values.
To run the Mixing Cycle process, select TUBE MIXING and/or RECIRCULATION.
Cycle Card Options
Customize the number of pumps and the duration according to your needs.
Adding New Cycle
If necessary, change the sequence of the steps by dragging the Grip Indicator up or down.
OPTIONAL: To repeat the sequence, fill in the number of repetition(s) in the REPEAT ALL option.
Then, tap RUN to execute the sequence.
If you encounter a problem during a run, perform a manual recovery using the Preloaded Files. This allows you to recover and re-run your samples.
To run the Preloaded Files:
Select one of these Preloaded Files according to your specific run.
Air Recovery: Draws in air to blow out the residual sample liquid from the transfer tubings and filter chip into the left (sample) tube.
Buffer Recovery: Uses the buffer liquid from the right (buffer) tube to push all the residual sample liquid from the transfer tubing and filter chip into the left (sample) tube.
Sample Recovery: Runs air recovery, buffer exchange, and a second air recovery for a more complete recovery.
Recirculation: Pumps liquid from the left (sample) tube over the membrane repeatedly to remove the sample bound to the membrane.
Note: The Priming file is for filling the chip with storage solution or initiating CONCENTRATE in a MANUAL PROFILE.
Tap OPEN.
Then, tap RUN.
