Although most imaging sessions are automated and executed according to the imaging schedule set in Rock Maker, it's possible to initiate an unscheduled imaging session from Rock Imager.
Using the Search sub-tab of the Plates tab, locate the plate in the imager.
Once you locate your plate, you have several options. Click on the plate to highlight it and select from the options below. If you want to image more than one plate, press and hold the CTRL key on your keyboard while selecting plates.
Note: All menu options are also available through right-click menus.
To image an entire plate with the MFI imaging settings, on the Imager menu, point to Image, click Plate, or click Plate Using to define a custom imaging setting.
To manually explore the contents of your plate, conduct a Manual Imaging session.
To image the current drop with the MFI imaging settings, on the Imager menu, point to Image, click Drop, or click Drop Using to define a custom imaging setting.
To image interesting drops, on the Imager menu, point to Image, click Interesting Drops.
To image interesting drops with a custom imaging setting, on the Imager menu, point to Image, click Interesting Drops Using.
When your plates have been imaged at least once, you can view and score your images in Rock Maker.
Place the plate in any available slot with the label facing towards you.
Tell Rock Imager to scan the barcode.
Choose from the following options:
To image an entire plate with the MFI imaging settings, on the Imager menu, point to Image, and click Plate(s).
To image an entire plate with the imaging setting(s) of choice, on the Imager menu, point to Image, and click Plate(s) Using. Rock Imager will display a dialog box that asks you to select available imaging settings.
To inspect your plates well-by-well or drop-by-drop without taking images, go to the Live Image tabThe Live Image tab is a sub-tab of the Imager tab. It is the tab that displays by default when you open the Imager tab. To find the Live Image tab, go to the Imager tab (top-left of your screen), and look for the tabs at the bottom-left of your screen. and use the manual navigation controls.
To image a specific drop with the MFI imaging settings, on the Imager menu, point to Image, and click Drop.
To image a specific drop with your choice of imaging settings, on the Imager menu, point to Image, and click Drop Using.
If you have tagged certain drops as interesting in Rock Maker, you can opt to image only those drops by going to the Imager menu and selecting Interesting Drop(s) in the Image menu to use the default imaging setting.
OR
Point to Image, and click Interesting Drop(s) Using to use a custom imaging setting.
When your plates have been imaged at least once, you can view the images in Rock Maker.
Tip: For imaging with Rock Imager 1000 and Rock Imager 2 MFI, use the following Camera Control and Optics Control settings as a starting point:
▶ Exposure: 50 ms.
▶ Gain: 5.
▶ Brightness: 100%.
