PULSE allows you to create a profile consisting of desired experiment settings for concentrating sample or performing diafiltration. You can set up a profile for the following experiment types:
On the Home Screen, open the EXPLORER menu and tap the ADD button in the lower-left corner of the EXPLORER window.
Type a name for your new profile by tapping on the pencil button.
Type the Starting Volume and Starting Concentration based on the sample you’re using, and then type the desired Final Volume. The Estimated Final Concentration will be automatically generated after you have set the other values.
OPTIONAL: If you need to concentrate more than 50 mL of sample, select ON in the >50 mL Volume option and type the volume in the Starting Volume in 2nd Tube option. The 1st tube is assumed to contain 50 mL.
In the Buffer Exchange option, select NONE if you want to purify the sample without exchanging buffer.
Select the correct chip valve in the Chip Valve option.
Select the Membrane Cutoff Size that matches the chip you’re using from the Membrane Cutoff Size drop-down list.
OPTIONAL: You can type some notes about your experiment in the NOTES field in the lower-right corner of your screen.
Tap SAVE.
On the Home Screen, open the EXPLORER menu and tap the ADD button in the lower-left corner of the EXPLORER window.
Type a name for your new profile by tapping the pencil button.

When performing buffer exchange you can not concentrate more than 50 mL, so you must leave >50 mL Volume OFF.
In the Buffer Exchange option,
Select Discrete to determine the diafiltration step or exchange count. For example, if you run 3 exchanges for 30 mL of buffer and start the buffer exchange at 20 mL, the PULSE will concentrate to 20 mL and then add 10 mL of buffer in 3 exchanges.
Select Continuous to diafiltrate continuously using a step size of 2 mL. For example, if you use 30 mL of buffer and start the buffer exchange at 25 mL, PULSE will concentrate to 25 mL and then add 2 mL of buffer, then concentrate back to 25 mL 15 times.
If you select Discrete, type the Exchange Count.
Type the value in the Sample Volume to Start option. This is the sample volume at which buffer exchange will begin.
Type the value in the Buffer Volume to Use option.
Type the total volume in the tube in the Total Buffer Volume option.
Select the correct chip valve in the Chip Valve option.
Select the Membrane Cutoff Size that matches the chip you’re using from the Membrane Cutoff Size drop-down list.
OPTIONAL: You can type some notes about your experiment in the NOTES field in the lower-right corner of your screen.
Tap SAVE.
You can configure more profile settings to meet your preferences by tapping the MORE button. There you can find a list of advanced settings that only apply to this profile, such as vacuum and pressure values, diaphragm timing, liquid recovery, and so on. Make sure that you understand these options before making any changes.
You can modify saved profiles in the EXPLORER window whenever you want to make changes to the existing profile settings, such as the starting volume, final concentration, final volume, and so on.
On the Home Screen, tap the EXPLORER menu, and then tap a profile you want to edit.
Tap the EDIT button and change the options you want to edit.
Tap SAVE.
