Iterative Screen Optimization (ISO)

ROCK MAKER allows you to automatically optimize a plate based on its prior scoring history. ISO can be performed in multiple rounds, with each new round optimizing the results from the round before it. During each round of optimization, the concentration of precipitants in your wells are increased or decreased by a percentage that you can set yourself. You can also use the default ROCK MAKER ISO percentage values, which are shown in the table below - they can be changed in the Manual Scoring Group under Setup. These values can also be changed for each round of ISO without changing them at the master level in the Manual Scoring Group.

For ISO, the ten default ROCK MAKER scores in the Default Manual Scoring Group are categorized in four groups: Clear (A), Crystal (B), Fine Precipitate (C), Heavy Precipitate (D). These four groups each have a unique formula to determine the adjustment in precipitate concentration during multiple rounds of ISO, but the step sizes used can be different for each of the ten score types, if desired. You can configure the step size for each score from the ROCK MAKER General Setup. Important: Any experiment you wish to optimize must be scored using the Manual Score Group. Only the score of the first drop in a well will be used for ISO.
