Random Layer¶
Random layer helps you design an experiment with several ingredient options. It randomly adjusts the weight, probability, pH, and concentration of selected ingredients in each well and its drop(s).

Allows you to select ingredient(s). Find out more about Creating a Random Layer. |
In a random layer, the ingredients in the Group panel are classified based on their types. Please see the image below.

Ingredient Distribution¶
ROCK MAKER uses the assigned probability in the Layers tab of the Layers Design panel to determine which ingredient groups to distribute to a well.
Groups with higher probabilities are more likely to be used in a well. In each selected group, ROCK MAKER chooses one or more ingredients to dispense in the well based on the weight value assigned to the ingredient in the Edit Ingredient panel.

Item |
Description |
---|---|
Name |
Displays the name of the selected ingredient. |
Low pH |
Allows you to select a stock with low pH. Note This field will appear if you select Buffer in the Type field. |
High pH |
This field is optional. It allows you to select a stock with high pH based on the low pH you select Note This field will appear if you select Buffer in the Type field. |
Type |
Displays the ingredient type. |
Concentration |
Allows you to define the start and stop concentration by typing the value in the Start and Stop fields. |
pH |
Displays the starting and stopping pH (see the Start and Stop fields) based on the selected Low pH and High pH. Note If you do not define the High pH, the Stop field will be N/A. |
Weight |
Allows you to define the weight of the ingredient you select. |
Concentration and pH Distribution¶
When designing a random layer, you need to define minimum and maximum values for the:
Concentration of each ingredient.
pH of each buffer ingredient.
ROCK MAKER uses the minimum and maximum values and grid steps number you can define in the Layers tab of the Layers Design panel to get sets of values to randomly design concentrations and pH levels in the random layer. The number of values is the number of steps you type in.
Each set is generated by defining numbers between the minimum and maximum values at a fixed increment determined by the number of grid steps. It results in a set of numbers for each attribute. The number of values in each set is equal to the number of grid steps.
Ingredient Variation¶
ROCK MAKER varies the ingredients by:
Determining which ingredient groups to use in each well based on the probability you assign to each group in the Group panel.
Randomly choosing one or more ingredients from each selected ingredient group based on the ingredient group’s probabilities.
Distributing the selected ingredients into the well.
Excluding Ingredients¶
Exclude List Item allows you to choose pairs of ingredients not to be mixed in random layer wells. As a result, none of the combinations of the Exclude List Item occurs.
